An FDA inspection conducted from April 17 to April 25, 2023, at Ciron Drugs and Pharmaceuticals Private Limited in Tarapur, India, resulted in a Form FDA 483 listing two significant observations related to Current Good Manufacturing Practices (cGMP).

The first observation highlighted inadequate validation of aseptic processes intended to prevent microbiological contamination of sterile drug products. Specifically, the firm's disinfectant efficacy study did not evaluate all critical surfaces within the aseptic cleanroom environment, such as certain RABS components, raising concerns about the comprehensive effectiveness of disinfection procedures.

The second observation concerned the insufficient testing and quality control of in-process bulk materials. The firm's bulk hold time studies for two injectable drug products were found deficient. These studies lacked proper documentation regarding the design and size of storage containers and the drug product volume-to-headspace ratio. Crucially, no stability evaluations were performed on the finished drug products manufactured from these bulk materials, and the firm lacked scientific justification for this omission.

In response to these findings, Ciron Drugs and Pharmaceuticals Private Limited is required to implement comprehensive corrective actions and provide a detailed response to the FDA addressing each observation to ensure full compliance with cGMP regulations.
